Description:

Grb10 (Growth factor receptor-bound protein 10, Maternally expressed gene 1) has two promoter regions, with two transcripts from each promoter. The alpha and delta transcripts arise from exon Un1 (exon 1a) in CpG island 1, whereas the beta1 and beta2 transcripts arise from exon Un2 (exon 1b) that overlaps a second CpG island (Arnaud P et al, 2003; Hikichi T et al, 2003).
The alpha and delta isoforms are maternally expressed in most tissues (Miyoshi N et al, 1998), whereas the beta isoforms are expressed only in the brain and from the paternal allele (Arnaud P et al, 2003; Hikichi T et al, 2003).
Biallelic expression was reported in brain but the assays used do not distinguish the isoforms (Hitchins MP et al, 2002).
Disruption of the maternal allele of Grb10 resulted in overgrowth of both the embryo and placenta such that mutant mice were approximately 30% larger than normal (Charalambous M et al, 2003). Grb10 is a candidate gene for the growth retardation of Silver-Russell syndrome .
A maternally methylated DMR has been well documented (Arnaud P et al, 2003; Hikichi T et al, 2003).
