Description:

The phenotypes associated with the A(iapy), A(vy) and A(hvy) mutant alleles of the agouti locus are dependent on the parental origin. These agouti mutants are attributable to integration of an intracisternal A-particle (IAP), which results in overexpression of the agouti protein. The overexpression of the A(iapy) allele is greater when maternally inherited, perhaps because of hypermethylation of the IAP element during spermatogenesis (Michaud EJ et al, 1994). The phenotypes of A(vy) and A(hvy) heterozygotes are affected by the parent-of-origin probably because of different degrees of methylation of the IAP element (Duhl DMJ et al, 1994; Argeson AC et al, 1996). A maternal epigenetic effect on A(vy) results from incomplete erasure of an epigenetic modification when a silenced Avy allele is passed through the female germ line (Morgan HD et al, 1999). The expression of the A(vy) allele is influenced by methyl supplementation of the maternal diet (Wolff GL et al, 1998).
